The lower in vitro chondrogenic potential of canine adipose tissue-derived mesenchymal stromal cells (MSC) compared to bone marrow-derived MSC is not improved by BMP-2 or BMP-6

Mesenchymal stromal cells (MSC) are used for cell-based treatment canine osteoarthritis (OA). Compared with human MSCs, detailed information on the functional characterisation of MSCs is limited. In particular, chondrogenic differentiation adipose tissue-derived (cAT-MSCs) challenging. this study, we aimed to compare cAT-MSCs bone marrow-derived (cBM-MSCs), focusing specifically their in vitro potential, or without morphogenetic proteins (BMP). cBM-MSCs and were characterised using flow cytometry reverse transcription-quantitative polymerase chain reaction (RT-qPCR). The potential all cMSC preparations presence TGF-β1 alone when supplemented 10, 100, 250 ng/mL BMP-2 BMP-6 was investigated RT-qPCR, biochemical, histochemical immunohistological analyses. Both expressed surface markers CD90, CD73, CD29, negative CD45 CD34, although expression CD73 CD271 varied donor tissue origin. Interestingly, ACAN SOX9 higher than cAT-MSCs. contrast cBM-MSCs, could not differentiate toward lineage BMP-2/-6, chondrogenesis inferior BMP-2/-6. Thus, have lower capacity cBM-MSC under studied culture conditions BMP-6. Therefore, further necessary explore OA treatments.
